The proposed mechanism is oral tolerance, a normal feature of gut immunology in which immune tissue in the small intestine samples intact proteins from food. Because recognizing a shape does not require bulk, the studied amount is 40 mg per day rather than the several grams typical of hydrolyzed collagen. The mechanism remains an active research question rather than a settled conclusion.
What the human studies measured
A randomized, double-blind, placebo-controlled trial in the Journal of the International Society of Sports Nutrition in 2013 followed healthy adults reporting knee discomfort after exercise. Over 120 days, investigators measured knee range of motion with a goniometer and how long participants could use a stepmill before joint discomfort began. A later multicenter randomized trial in Nutrition Journal followed adults with persistent knee discomfort for 180 days at the same 40 mg intake, using standardized joint questionnaires.

- Walk regularly, since cartilage relies on movement to circulate joint fluid
- Keep body weight in a healthy range to reduce load through the knee
- Build strength in the muscles around the hip and thigh
- Vary sitting positions rather than holding deep flexion
- Increase training load gradually

UC-II® products are typically one small capsule daily, often in the evening away from a heavy meal, since the mechanism depends on the molecule reaching intestinal immune tissue intact. The published studies ran three to six months, which is the timescale the evidence reflects. It comes from chicken cartilage, relevant for anyone avoiding poultry-derived ingredients.
